氮源浓度对黄原胶流加发酵过程的影响

摘要: <正>引言 黄原胶作为最重要的微生物多糖在食品、采油等工业中得到广泛应用。黄原胶生产过程的经济性取决于黄原胶发酵的最终浓度,该浓度与生物反应器中诸多环境因素密切相关,其中氮源的种类及浓度对发酵过程均有重要影响,主要是影响细胞生长并通过细胞量的变化影响黄原胶合成。氮源可分为有机氮源和无机氮源,早期研究表明有机氮源要优于无机氮源,常用DDS(Distiller’s Dried Solubles)、CSL(Corn Steep Lique)、蛋白胨等。对浓度影响的研究也主要是针对有机氮源,但有机氮源组成复杂,常含某些未知因子影响细胞生长。此外,有机氮源还不利于黄原胶的分离回收,因此以组成确定的无机氮源考察氮源浓度对发酵过程的影响更好。

Abstract: Using (NH4)2SO4 as the N - source, the effect of the initial N - source concentration on xanthan fed - batch fermentation was studied. The best concentration was about 4 gdddddddddddL-1. This result was compared with previous studies. The oxygen limitation and the stagnant zone might be the main reason for the decrease of the production rate in high cell concentration. By improving the oxygen supply capacity of the equipment, optimizing the N - source concentration and fed- batch control of glucose concentration, xanthan concentration achieved 40 gddddddddddL-1 in 55 hours.
