EFFECT OF ADSORPTION DENSITY ON PORE DIFFUSIVITY OF PROTEINS IN ION EXCHANGER

Abstract: The ion-exchange adsorption kinetics of bovine serum albumin (BSA) and γ-globulin into an anion exchanger,DEAE-Spherodex M, has been studied by batch adsorption experiments.The pore diffusion model was used to predict pore diffusivity.The effect of initial protein concentration and average adsorption density on pore diffusivity was investigated.The results showed that pore diffusivity decreased exponentially with increasing initial protein concentration and average adsorption density. Moreover, the larger the dimension of the protein, the greater the decline of pore diffusivity was observed.

摘要: 通过间歇吸附动力学实验,采用孔扩散模型研究了牛血清白蛋白和γ-球蛋白在阴离子交换剂中的扩散行为,考察了蛋白质初浓度和平均吸附密度对孔扩散系数的影响.结果表明,随蛋白质初浓度和平均吸附密度增大,孔扩散系数均呈指数下降,且蛋白质分子尺寸越大下降趋势越明显.
