Abstract:
Rhodococcus sp. CCZU10-1 has been isolated from soil samples with high nitrilase activity against 3-cyanopyridine using the enrichment culture technique. The preferred carbon sources,nitrogen sources,and inducer were glucose(10 g/L),a composite of peptone(10 g/L)and yeast extract(5 g/L),and ε-caprolactam(2.0 mmol/L),respectively. The optimal reaction temperature and pH for 3-cyanopyridine-hydrolyzing activity were 30 ℃ and pH 7.0,respectively. Using fed-batch method,a total of 927 mmol/L nicotinic acid has been accumulated in the reaction mixture after the 10th feed.

利用富集培养技术筛选获得一株高活性烟腈水解菌株Rhodococcus sp. CCZU10-1。经研究,最适产酶培养条件和催化反应条件为:碳源为葡萄糖(10 g/L),氮源为酵母蛋白胨(10 g/L)与酵母膏(5 g/L),诱导剂ε-己内酰胺(2.0 mmol/L),培养温度为30 ℃,初始培养pH值为7.0,最适催化反应温度和pH值分别为30 ℃和7.0。分批补料烟腈10批,烟酸的累计浓度为927 mmol/L。
