中空纤维亲和膜吸附蛋白质的动力学模型

摘要: 通过对蛋白质溶液在中空纤维亲和膜中流动状况和吸附反应的分析,建立了中空纤维亲和膜蛋白质吸附的动力学表达式.以人γ─免疫球蛋白在A蛋白为配基的亲和膜上的吸附为例,分析了所有亲和吸附位点的反应速度常数都一致的均一吸附和存在两种亲和吸附位点、反应常数不同的非均一吸附两种情况并建立了相应的模型.用有限差分法获得了吸附透过曲线并与实验结果进行了对照.结果表明非均一吸附模型的预测与实验值更为接近.

Abstract: The expression of process kinetics of protein adsorption in a hollow fiber affinity membrane has been established through analysis of the flow distribution and adsorption reaction.The process of human Y-immunoglobulin adsorption onto an immobilized protein A hollow fiber membrane was taken as an example.Homogeneous binding of uniform sites with the same reaction constant and heterogeneous binding of two different sites with different reaction constants have been considered.Breakthrough curves are obtained through numerical solution of the partial differential equations by finite difference method.The comparison between the predicted curves and the experimental curves showed a better agreement for the heterogeneous binding model.
